Ractopamine is a drug that is used as a feed additive to promote leanness in animals raised for their meat. Pharmacologically, it is a beta-adrenoceptor agonist. It is the active ingredient in products known as Paylean for swine and Optaflexx for cattle, developed by Elanco Animal Health, a division of Eli Lilly and Company, for use in food animals for growth promotion.

Ractopamine has been banned in the European Union, Taiwan, mainland China and Russia.

As you know from the news: Rosselkhoznadzor on February 11, 2013. introduces prohibitions supply frozen meat and meat products from the United States. This is due to the fact that the American Veterinary Service does not observe the rules of ractopamine. Supply of chilled meat for that reason are prohibited from February 4. The use of ractopamine in Russia is prohibited. Rosselkhoznadzor notified the case management Safety and Inspection Service Food USDA.

1. Extraction by ASE

2. Automated On-Line SPE-LC for HPLC Sample Preparation and Automation.

Solid-phase extraction (SPE) is a technique that is commonly used in laboratories to isolate the analytes of interest from complex matrices.

Example of analysis of pork:

Sample preparation: Extract 5,0 g pork chop with approx.. 50 ml of 4:1 acetonitrile : isopropanol using ASE extractor. Chill to precipitate fat, and filter.

In this example, we extracted pork with organic solvents in an Accelerated Solvent Extractor. Evaporation and reconstitution proved troublesome, so we avoided that by using solid-phase extraction (SPE).

SPE is most effective when it is orthogonal to the analytical separation, so we used cation exchange to retain the drugs, and washed away lipids and other interferences with organic solvent. The auxiliary valve then transferred the drugs to an Acclaim C30 column for RP-HPLC analysis.
